Path length correction

For many assays such as DNA quantification and protein determination, users frequently compare absorbance results from a microplate reader with those from a 1 cm path length cuvette spectrophotometer.

All microplate readers from BMG LABTECH that are capable of reading absorbance have a 1 cm path length correction built-in via an internal fitting algorithm. Using this correction technique, the 1 cm data from a microplate assay is given with no calculations or estimations made by the user. The path length correction feature can be turned on or off in the software as your needs dictate. Waterpeak-based path length correction can further be used to compensate for well-to-well variation in the filling volume and can compensate for meniscus effects as explained in the HowTo Note: How to deal with path length and meniscus in microplates.

A linear regression comparison of data from a BMG LABTECH microplate reader and a 1 cm cuvette spectrophotometer produces an r² value of over 0.99 in the UV and visible portions of the spectrum.
